Dimethyl fumarate (DMF) is the methyl ester of fumaric acid and is named after the earth smoke plant (Fumaria officinalis). DMF combined with three other fumaric acid esters (FAEs) is solely licensed in Germany as an oral therapy for psoriasis (trade name Fumaderm®). Since 2013, it has been used to treat adults with relapsing multiple sclerosis (trade name Tecfidera®). In 2017, a new oral formulation of DMF (trade name Skilarence®) was approved by the European Medicines Agency (EMA) for use in Europe as a treatment for moderate-to-severe plaque psoriasis. DMF is thought to have immunomodulatory properties without causing significant immunosuppression.

Synthesis and application of coumarin type dye sensitizer

The invention relates to a coumarin functional dye containing a thiophene bridge chain in the field of fine chemical industry and organic photoelectric material applications. The structure of the coumarin functional dye takes coumarin and a derivative thereof as an electron donor, contains a thiophene structure unit capable of adjusting an absorption spectrum and a fluorescence emission spectrum as the bridge chain and is further connected with a cyanoacetic acid electron withdrawing group. Coumarin-thiophene, POCl3 / DMF (dimethyl fumarate) are added into a reaction container by adopting general reaction for reaction so as to get a 5-(7-substiutted-2-carbonyl-2H-benzopyran-3-yl) thiophene-2-formaldehyde intermediate (II) with an aldehyde group; and the intermediate II with the aldehyde group further reacts with cyanoacetic acid to get the coumarin dye connected by the thiophene. As the coumarin is taken as a chromophore, the electron donating capability is good; the thiophene has high electron cloud density and special optical properties and electron transmission capability; and the electron withdrawing group of the cyanoacetic acid is further connected for enabling the dye to have good light, thermal and chemical stability and photoelectric properties. Therefore, the dye can be used as a photosensitive dye for dye-sensitized solar cells.

Continuous production process for dimethyl fumarate

The invention discloses a continuous production process for dimethyl fumarate. The process comprises the following steps of: uniformly filling a strong acid cation exchange resin catalyst into a reaction cylinder, and heating at the temperature of between 60 and 100 DEG C; adding fumaric acid and a methanol solution which serve as reaction raw materials into the reaction cylinder from an inlet of the fixed reaction cylinder, regulating the flow velocity of a flow pump to ensure that the raw materials are kept in the reaction cylinder for 4 to 8 hours, and collecting a reaction product at an outlet; introducing the reaction product into cold water, stirring, and crystallizing a large number of white crystals; filtering, and washing a filter cake by using 8 to 10 percent Na2CO3; and filtering again, washing the filter cake to be neutral by using distilled water, drying at room temperature, and thus obtaining the dimethyl fumarate. In the process, strong acid cation exchange resin is utilized, does not corrode reaction equipment and pollute the environment, can be repeatedly used and is a good environment-friendly catalyst. The catalyst is fixed in the reaction cylinder; the materials are continuously fed and discharged, so that the continuous operation of the reaction can be realized; and the process has simple reaction conditions and makes the operation convenient.

Novel method for enhancing efficiency of human T cells infected by slow-viruses carrying CAR (chimeric antigene receptor) genes

The invention relates to a novel method for enhancing the efficiency of infecting human T cells by slow viruses carrying CAR (chimeric antigene receptor) genes. A CAR-T technology is regarded as a technology for most possibly curing tumors; although the technology has breakthrough, bottleneck problems still existing in the aspect of preparation of CAR-T cells cannot be solved, for example, the innate immune response of T cells can enable the efficiency of infecting the T cells by the slow viruses carrying CAR genes to be low, so that the CAR-T cells are low in preparation efficiency and high in cost. For the problem of improving the efficiency of infecting the T cells by the slow viruses carrying CAR genes, the applicant conducts a series of experiments and improvement and discovers that the infection efficiency of the CAR slow viruses can be remarkably improved when the T cells are treated by DMF (dimethyl fumarate). The novel method has guiding significance and application values inthe aspects of greatly improving the preparation efficiency of the CAR-T cells, shortening the in vitro amplification time of the CAR-T cells, lowering the production cost of a CAR-T therapeutic method and the like.
