The invention relates to a construction method for obtaining an aseptic
seedling of a
lotus flower, and belongs to the technical field of
plant tissue culture. The construction method comprises the following steps of (1), sterilization: selecting a round and full
lotus seed of a Guangchang
white lotus root, and disinfecting by using 75 percent
alcohol and 3 percent
sodium hypochlorite on an aseptic operating platform for later use; (2),
hull breakage and soaking: breaking a
hull by using
pruning shears, leveling and soaking the seed of the
lotus flower, whose
hull is already broken, in asepticwater for 7h or above, wherein the seed absorbs water and expands, and a seed
husk is separated from a seed leaf; (3), stripping of the seed
husk: stripping a seed coat outside the seed, soaking theseed without the seed
husk in the 75 percent
alcohol for 2min for disinfection, taking the seed out, and air-
drying; (4): obtaining of a seed
embryo: manually pinching open the seed, which already absorbs the water and expands, after
moisture on the surface of the seed is evaporated, and taking the seed
embryo down by using
tweezers; (5), primary selection and
inoculation: carrying out the primaryselection on the
embryo, and inoculating the embryo which is qualified in the primary selection in an MS (Murashige-Skoog) culture medium. The construction method is used for obviously improving theutilization rate and
survival rate of the lotus seed, and moreover, the
contamination rate and the
loss rate are furthest decreased.
