Terahertz modulator based on silicon-based microstructure on SOI, system and method

The invention provides a Terahertz modulator based on a silicon-based microstructure on SOI, a manufacturing method and a modulation system. The Terahertz modulator successively comprises an Al2O3 substrate of a bottom layer, a SiO2 isolation layer, the silicon-based microstructures, and an Al2O3 passivation layer from bottom to top. The silicon-based microstructures are periodically arranged on the SiO2 isolation layer. Each silicon-based microstructure comprises a two-layer square Si-based step structure. The modulation system comprises a semiconductor laser, a laser modulator, the Terahertzmodulator, a Terahertz radiation source, and a Terahertz detector. In the invention, a reflectivity which is lower than 22% is achieved for a Terahertz wave of 0.4 THz to 0.85 THz and a minimum of 18% can be reached at 0.82 THz so that the reflectivity of a modulation device to the Terahertz wave can be significantly reduced and a utilization rate of the Terahertz wave is improved. A 64.5% modulation depth can be achieved under 808 nm laser radiation with 1200 mw power. Compared with a traditional silicon-based Terahertz modulator, a Terahertz imaging diffusion area can effectively increase aresolution and make the resolution reach above 21.9% in an imaging system.

Purification method of semaglutide

The invention relates to a purification method of semaglutide. The method comprises the following steps that 1) cracking resin prepared through a solid-phase synthesis method and semaglutide crude peptide from which a protecting group is removed are obtained, and the semaglutide crude peptide is dissolved by using a phosphate buffer solution containing an organic solvent; 2) a semaglutide crude peptide solution is filtered by using an organic filter membrane of 0.22 micro meter, and a filtrate is collected for later use; 3) in first-step purification, by taking a reversed-phase filler as a stationary phase, a phosphate buffer solution containing an organic solvent as a phase A and acetonitrile as a phase B, gradient elution is conducted; 4) in second-step purification, by taking a reversed-phase filler as a stationary phase, a dilute acetic acid solution as a phase A and acetonitrile as a phase B, gradient elution is conducted; and 5) freeze drying is conducted on a concentrated sampleto obtain the semaglutide. According to the method, the phosphate buffer solution containing the organic solvent is used for dissolving the crude peptide, and the pH value of the dissolved crude peptide solution is more stable and crude peptide is not prone to separate out, so that the blockage of a chromatographic column and equipment is avoided, and the separation effect is improved.

The invention provides a method for semi-quantitative determination of ethyl carbamate through a thin layer chromatography. According to the method, mixed liquor of cinnamyl aldehyde and phosphoric acid is used as a fluorescence derivating agent, mixed liquor of methyl tertiary butyl ether, petroleum ether and methyl alcohol is used as mobile phase developing solvent, a chromatographic sheet is used as an immobile phase, after the ethyl carbamate developed by a thin layer is reacted with the fluorescence derivating agent, accordingly, green fluorescent spots can be observed at the position of 365 nm through a common ultraviolet analysis meter, the Rf value is 0.85, and the lowest drug loading capacity of a thin layer plate is 50 ng of ethyl carbamate. According to the limiting standard of 5 mg/L ethyl carbamate in wine, 2.5 mg/L EC can be detected directly, and the sample application amount of the thin layer plate is generally 20 microliters maximally. The method is small in number of needed samples, rapid, high in sensitivity, simple in step, small in number of needed equipment and easy to operate. In accordance with the principle, a rapid test kit can be researched and developed, and the method is used for rapidly detecting the ethyl carbamate in alcoholic beverages and fermented food.
