The present invention relates to a novel nasal formulation for the treatment and prophylaxis of nasal congestion. When an infant is born, its nasal passages are relatively sterile, in time the nasal passages are colonized by fungi, molds and other organisms which compromise the integrity of the nasal and paranasal mucosa. The presence of these organisms elicits a mild chronic inflammatory reaction of the nasal and paranasal mucosa, in some cases leading to congestion and excess production of mucus. This chronic inflammatory process compromises the integrity of the nasal and paranasal mucosa, making it more vulnerable to binding with allergens and other invading organisms such as the rhinovirus. The purpose of this invention is to establish a treatment and prophylaxis of nasal congestion, which will control the presence of the invading organisms in the nasal and paranasal mucosa, reducing congestion as well as mucus production. By reducing the mild chronic inflammation in the nasal passages, prophylaxis with this nasal formulation passively reestablishes the integrity of the nasal and paranasal mucosa, thereby reinforcing the body's defenses against the implantation of allergens and of organisms such as the rhinovirus.

A method for recombinantly expressing a macromolecule in a host cell is disclosed which involves culturing a host cell which contains two nucleic acid sequences, i.e., a first nucleic acid sequence encoding a membrane-permeabilizing agent and a second nucleic acid sequence encoding a desired macromolecule under the operative control of an inducible promoter, to a selected cell density that permits accumulation of the agent. The host cell thereby allows transport through the membrane of small molecular weight compounds. These resulting host cells are cultured in the presence of a nutrient cocktail that contains components that can transport through the disrupted cell membrane, e.g., an inducing agent that induces the tightly regulated promoter and metabolic requirements that permit expression of the macromolecule.

With only 19 amino acids, Duramycin is the smallest known polypeptide that has a defined 3-dimmensional binding structure. Duramycin binds Phosphatidylethanolamine (PtdE) at a 1:1 ratio with high affinity and exclusive specificity. As an abundant binding target, PtdE is a major phospholipid and accounts for about 20 % of the phospholipid content in mammalian cellular membranes. PtdE is externalized to the surface of apoptotic cells, and also becomes accessible in necrotic cells due to compromised plasma membrane integrity. Given the unique physicochemical properties of Duramycin and the availability of PtdE in acute cell death, the goal of this study is to develop and evaluate 99mTc-HYNIC-Duramycin as a novel molecular probe for imaging PtdE. 99mTc-HYNIC-Duramycin is a low-molecular weight, fast-clearing radiopharmaceutical that detects apoptosis / necrosis by binding to PtdE. The goal was to quantify the uptake of 99mTc-HYNIC-Duramycin in the area-at-risk after myocardial ischemia and reperfusion, and to determine the window of detection.
