The present invention is directed to recombinant human antibodies specific for Cn2 toxin from C. noxius scorpion venom. The antibodies are able to recognize the toxin and preferably neutralize it as well as the whole venom of C. noxius scorpion. This invention is also directed to a human non-immune phage display library. One clone that specifically binds the Cn2 toxin was affinity matured by directed evolution. Three cycles of maturation were performed and several scFv clones were isolated which specifically recognize toxin Cn2 with increased Kd of 446 fold. All variants were monomeric and only variants 6009F, 6105F and 6103E showed to be capable of neutralizing toxin Cn2 and the whole venom. Variant 6009F recognizes a different epitope than that of BCF2, a murine monoclonal antibody raised against scorpion toxin Cn2 which is also capable of neutralizing both Cn2 toxin and the whole venom when tested in mice, as well as that of commercially available polyclonal antibody fragments antivenom from horse. The scFv 6009F is the first reported recombinant human antibody fragment capable of neutralizing a scorpion venom. These results pave the way for the generation of safer autologous recombinant neutralizing antivenom against scorpion stings. The antibodies of the present invention can be used as part of a composition to treat those in need of treatment including those already stung by one or more scorpions, particularly C. noxius scorpions.

The invention belongs to the field of research and development of polypeptide medicines and discloses a scorpion venom heat-resistant synthetic peptide and application thereof. An amino acid sequence of an SVHRP (scorpion venom heat-resistant peptide) is detected out from BmK (Buthus martensii Karsch) which is a traditional Chinese medicine; according to animal experiment verification, a scorpion venom heat-resistant peptide extract liquid sample has pharmacological activity in prevention and treatment of intractable epilepsy, Parkinson's disease and Alzheimer's disease, the sample is subjected to LaGm composite and repeated fast magnetic separation prior to nanoLC-ESI-MS (nano-liter reversed-phase chromatography and electrospray ionization mass spectrometry) integrated mass spectrometry parallel experiment to detect out a polypeptide sequence formed by 15 amino acid residues. The scorpion venom heat-resistant synthetic peptide is prepared by solid-phase chemical synthesis, chromatography purification and mass spectrometry identification. An amino acid sequence of the scorpion venom heat-resistant synthetic peptide is as shown in SEQ ID NO.1 and keeps pharmacological activity and safety of the scorpion venom heat-resistant peptide, and the scorpion venom heat-resistant peptide also has a characteristic of biological activity in promotion of reverse differentiation of neuroglial cells into neural stem cells.

Method for processing scorpion

The invention relates to a method for processing Chinese medicinal materials, particularly to a method for processing scorpions. The prior method for processing the scorpions is carried out by boiling, thereby inducing the live scorpions to secrete scorpion venom to the exterior after suffering from heat flash stimulation, and the scorpion venom is thermolabile and soluble in water, so as to lose a large amount of effective ingredients at the beginning of the processing. The invention provides a novel method for processing the scorpions, which comprises the following steps of soaking a live scorpion in clean water so as to cause the scorpion to discharge food and residues in digestive tract out of the body under a comfortable environment, and killing scorpion by refrigerating at low temperature so as not to loss effective ingredients in the scorpion. The method can maximally retain the effective ingredients of the scorpion. The obtained scorpion by the method has the peptide and protein extraction amount of 1.6-2.0g/3g scorpion, and extraction rate of 53.3-66.6%, which is increased by a factor of two compared with the prior process. The inventive processing method has the advantages of improved therapeutic effect of scorpion, reduced consumption of raw materials, and increased benefit.
