The method comprises the following steps: taking smoke from a measuring hole at the desulphuration outlet by using a smoke sampling gun with a built-in glass fiber filter membrane; taking down the glass fiber filter membrane in the smoke sampling gun, and acquiring the total crystal water content m<2> in a gypsum sample; acquiring the molar weight of total CaSO4 by a 'sulfur trioxide determining-barium sulphate gravimetric method' in section 11 in GB / T5485-2012 and a formula; acquiring the mass X1 of CaSO4.2H20 in the gypsum sample by a linear equation in two unknowns; calculating the content of the gypsum particles in the smoke; and verifying whether the data are qualified or not. The method has the advantage that the method for detecting the content of the gypsum particles in the smoke of the desulphuration outlet is simple, practical, rapid and accurate.
