The application provides a separation and purification method of
aesculin,
aesculetin and
aesculin in
cortex fraxini, which comprises the following steps: firstly, heating extraction is performed on the crushed
cortex fraxini raw material; secondly, plate and frame pressure
filtration is performed on the obtained extract, and the filtrate is subjected to
chromatographic separation on a
macroporous resin column; then, 10-30%
ethanol is used for resolving the
macroporous resin column and purifying
aesculin; subsequently, 50-80%
ethanol is used for resolving the
macroporous resin column, and the resolving liquid is subjected to enzymolysis and
aesculetin purification; finally, the enzymolysis
mother liquor is treated and aesculin is purified. The multi-layer separation and purification scheme can simultaneously and efficiently separate and purify the three high-value
active components of aesculin,
aesculetin and aesculin in
cortex fraxini, greatly improves the comprehensive
utilization rate of cortex fraxini resources, creates higher
product value, reduces the use amount of organic solvents, reduces the production cost, reduces the
pollution to the environment, and meets the green and
environmental protection production concept.
