Method for enhancing high GC content DNA fragment amplification efficiency by organic solvent

The invention relates to a method for strengthening the amplification efficiency of DNA segments with high GC content through an organic solvent, which comprises the following steps: firstly, extraction of DNA to be amplified; secondly, configuration and optimization of a PCR system; thirdly, setting and operation of PCR conditions; and fourthly, electrophoretic detection of PCR products, wherein the optimization of the PCR system is to add the organic solvent into the PCR reaction system; and the electrophoretic detection of the PCR products is to adopt 1 percent of sepharose gel, to dye by an EB liquid after glue leaking, to use an ultraviolet light-splitting transmissometer to observe the position of amplified segments in the gel, and to compare the amplified segments with corresponding molecular weight standards. The method has obvious effect of amplifying the DNA segments which are rich in GC alkaline base, so that the amplification of sequences with high GC content is more efficiently and has more purposefulness. The method has important theoretical significance and application value on development of the molecular biological technical tests.

Method of atomic transformation

This invention provides a method for atomic transformations carried out under conditions akin to chemical catalysis. Liquid and solid state catalysts are used in a two-step process. We have found that the high ionic / electric activity of concentrated sodium hydroxide solution in combination with heating is sufficient to induce atomic transformation and provide a solid phase catalyst of high aluminum and silicon content. This product when heated at a temperature of 1000° C. yields numerous elements of higher atomic masses. The method comprises the following steps: firstly, standing a mixture and observing whether the mixture has a solvent layer or not; if so, testing a closed cup flash point of the solvent layer; if a measured closed cup flash point value is more than 60 DEG C, determining that the mixture is a common chemical; if the measured closed cup flash point value is smaller than 60 DEG C, determining that the mixture is flammable liquid; secondly, combusting metal powder without the solvent layer; testing a solid combustion speed of the metal powder; if a measured combustion speed value is more than 2.2mm / s, determining that the metal powder is a flammable solid; then, measuring a closed cup flash point of a mixture with the measured combustion speed value smaller than 2.2mm / s; if the measured closed cup flash point value is more than 60 DEG C, determining that the mixture is the common chemical; finally, continuously combusting the mixture with the closed cup flash point value smaller than 60 DEG C and observing whether the mixture is continuously combusted or not; if so, determining that the mixture is the flammable solid; otherwise, determining that the mixture is the common chemical. The method disclosed by the invention has the advantages of reliable evaluation results and stable utilization conditions.

The delay time temperature coefficient (T[CD]) of the prepared piezoelectric ceramic is approximate to zero, the surface acoustic wave propagation velocity (Upsilon[s]) is 2500 m / s or so, and the dielectric constant is 230 or so. The mechanical factor of merit (Q[m]) is 2000 or so, the surface acoustic wave electromechanical coupling factor (K[s]) is 0.18 or so, and the Curie temperature is 350 DEG C or so.
