The invention discloses a method for detecting a breast duct of a root tissue of a kok-saghyz plant by a freezing slide preparation technology. The method comprises the following steps: selecting a complete plant tissue of the kok-saghyz to be detected, and putting into a stationary liquid; performing vacuum air exhaust on the stationary liquid, wherein the air exhaust time is controlled to be 1-3 min; placing at 4 DEG C for 6-8 hours; taking out the plant tissue in the stationary liquid and performing immersion cleaning for three times, wherein each time is 30 min; the plant tissues are dehydrated step by step through a sucrose solution; after the dehydrated plant tissue adsorbs excessive moisture, embedding the dehydrated plant tissue by using a Biosharp embedding agent, placing the plant tissue at -20 DEG C for 30-45 minutes, slicing the root tissue by using a freezing slicer, adding 10% glycerol for sealing, and solidifying and sealing by using the glycerol for 10 minutes to obtain a slide specimen so as to obtain the distribution condition of the breast ducts in the kok-saghyz root tissue. According to the method, the distribution and quantity conditions of the breast ducts in the kok-saghyz roots can be rapidly detected, the detection period is greatly shortened, and the growth conditions of the breast ducts of the kok-saghyz can be intuitively reflected.
