The method comprises the following steps of purifying a preparedsample through an efficient gel column after the sample is introduced, cutting part of fraction flowing out of a component to be measured, diluting the cut fraction by using a dilution pump to reducethe solvent strength of the fraction, mixing the fraction by using a mixer, and enriching the fraction by using a solid-phase extraction column; and after the sample is enriched, eluting the enrichedcomponent from the flow direction opposite to the sample enrichment by using a mobile phase of an analysis pump through valve switching, and detecting and measuring the contents of the benzo [a] pyrene and the metabolite thereof through ultra-high performance liquid chromatography column separation and a fluorescence detector. According to the method, efficient purification and solid-phase extraction enrichment of the sample can be realized at the same time, the analysis sensitivity of the method is greatly improved, the pretreatment of the analysis sample is greatly simplified, and the contents of the benzo [a] pyrene and the metabolite thereof in the order of ng/mL in a blood and urine sample can be accurately measured. A sensitive, accurate and reliable novel method is provided for research on the correlation between smoking and health.
