The invention further discloses suspension fluid prepared by using the suspending agent. The coccidiosis oocyst cryoprotectant comprises the following components: sodium dodecylsulfate, distearoyl phosphatidyl ethanolamine-polyethylene glycol, tartaric acid, ammonium sulfate, sucrose, fructose, sodium dihydrogen phosphate, disodium hydrogen phosphate, a standard PBS buffer solution and water for injection. The cryoprotectant is reasonable in formula, and can effectively inhibit ice crystal recrystallization in the cryopreservation process, regulate and control ice crystal morphology, reduce ice crystal growth rate and decrease damage of ice crystals to coccidiosis oocysts; and meanwhile, phospholipid molecules can be adsorbed to the walls of the coccidiosis oocysts,so the ice damage resistance of the coccidiosis oocysts is improved, and the coccidiosis oocysts can be stored in liquid nitrogen for a long time. Compared with a traditional cryoprotectant, the coccidiosis oocyst cryoprotectant of the invention does not contain toxic molecules such as DMSO and components such as expensive serum, avoids the toxic damage of DMSO to coccidiosis oocysts, lowers production cost, has effectively improved applicability in the field of coccidiosis vaccines is enhanced, and the cryoprotectant and is beneficial for wide popularization and application.

The invention relates to biological immunity and a vaccine, and in particular discloses a coccidiosis vaccine component having a cross immunological protection capacity and a vaccine containing the coccidiosis vaccine component. The coccidiosis vaccine component is a transgenic coccidiosis strain capable of expressing heterogeneous coccidiosis immunity dominant antigens. The transgenic coccidiosis strain, which simultaneously has immunogenicities of a parental coccidiosis strain and a heterogeneous coccidiosis strain, can enhance the cross immunological protection capacity of the coccidiosis vaccine component when the transgenic coccidiosis strain serves as the coccidiosis vaccine component. Based upon experimental studies, it discovers that the cross immunological protection capacity, which is provided by the novel vaccine component, is positively correlated to the quantity of the expressed heterogeneous dominant antigens; therefore, preferably, a plurality of immunity dominant antigens are expressed. According to the coccidiosis vaccine component and a construction strategy thereof provided by the invention, the immunological protection capacity can be offered for various insect infections; and the cross immunological protection capacity is universal.

The use method comprises the steps of: mixing one, two or more than two kinds of cellulose ether with lowest critical solution temperature or a mixture consisting of the cellulose ether with high-temperature water; pouring the high-temperature water containing the cellulose ether into low-temperature water and stirring; dissolving the cellulose ether or the mixture rapidly; and using the dissolved cellulose ether or mixture as the chicken coccidiosis vaccine suspension. The preparation method is as follows: adding the suspension prepared by the chicken coccidiosis vaccine suspending agent into chicken coccidiosis tetravalent living vaccine and then mixing. The use method and the preparation method have the following advantages: (1) the suspending agent disclosed by the invention can be dissolved into water rapidly; (2) the prepared suspension can keep the oocyst of the chicken coccidiosis living vaccine from settling into water within 8 hours, and the uniformity of the number of the vaccine oocyst which is driven by chicken is increased; (3) chicken flocks can be immune without cutting off water, and water cut-off stress is decreased; (4) the chicken flocks can be also immune by cutting off water, and immune time is shortened; and (5) the side effect of the vaccine can be reduced.

Resistance index estimation method for selective breeding of chicken eimeria tenella resistant individuals

The invention relates to the field of poultry breeding and specifically relates to a method for accurately estimating each index concentration after challenge of chicken eimeria tenella according to resistance indexes measured after immunization with a chicken coccidiosis vaccine. The method comprises the steps of immunizing samples to be screened with the chicken coccidiosis vaccine and then measuring five resistance indexes in the blood plasma, namely CAT, SOD, GSH-Px, MDA and IFN-gamma, and then accurately estimating the corresponding concentration of each index after the challenge (infection) of the chicken eimeria tenella by use of a pre-established regression equation of each resistance index according to the five resistance indexes measured after immunization with the chicken coccidiosis vaccine so as to select chicken eimeria tenella resistant individuals by use of the five resistance indexes under the condition of non-challenge (infection), wherein the regression equation of the resistance index refers to the quantitative relation of the resistance index after immunization with the chicken coccidiosis vaccine and challenge (infection). The method is capable of realizing conventional selective breeding of the chicken coccidiosis, and is safe and significant.
