The invention discloses a method for performing
induced mutation on
griseofulvin strain through
diethyl sulfate and
ultraviolet rays. The method comprises the following steps: firstly, crushing selected
bacterial colony through glass balls, preparing
spore suspension, sucking 1ml of single
spore suspension, diluting to obtain a contrast, and adding filtrate to double discs in which a
diethyl sulfate containing culture medium is prepared in advance; uniformly irradiating for 30mn through the
ultraviolet rays, covering with a black cloth, diluting with
sterile water, uniformly applying onto a flat vessel to obtain a single
bacterial colony, transferring the single
bacterial colony into a constant-temperature box for culturing, selecting a mitogenetic single bacterial colony 8 days after thebacterial colony grows, and transferring the mitogenetic single bacterial colony into a bevel for culturing for 8 days; transferring the
manure bacterial colony into a
shake flask for primarily screening, selecting 10 bevels of which the
potency unit is 3% more than the beginning bacterial colony, transferring the bevel, culturing until the bacterial colony is
manure, and screening again through the
shake flask; selecting 5 bevels with relatively high unit after screening again, burying the bevels with corresponding numbers into sandy soil, and storing. With the adoption of the method, the fermenting unit can be further increased; and the strain can remain outstanding properties.
