The present invention relates to protein matrix materials and devices and the methods of making and using protein matrix materials and devices. More specifically the present invention relates to protein matrix materials and devices that may be utilized for various medical applications including, but not limited to, drug delivery devices for the controlled release of pharmacologically active agents, encapsulated or coated stent devices, vessels, tubular grafts, vascular grafts, wound healing devices including protein matrix suture material and meshes, skin/bone/tissue grafts, biocompatible electricity conducting matrices, clear protein matrices, protein matrix adhesion prevention barriers, cell scaffolding and other biocompatible protein matrix devices. Furthermore, the present invention relates to protein matrix materials and devices made by forming a film comprising one or more biodegradable protein materials, one or more biocompatible solvents and optionally one or more pharmacologically active agents. The film is then partially dried, rolled or otherwise shaped, and then compressed to form the desired protein matrix device.

Bioprosthetic devices include an exterior biological tissue member which at least partly defines a cavity, and a proteinaceous biopolymer which fills the cavity, and intercalates and is chemically bound (fixed) to the tissue of the surrounding biological tissue member. In preferred forms. the bioprosthetic device is a bioprosthetic vertebral disc having a fibrillar outer annulus which surrounds and defines an interior cavity and is formed by removal of at least a substantial portion of the natural gelatinous core therefrom. A surface which carries a material which is infected with prions is cleaned with an alkaline cleaning solution to remove as much proteinaceous material as possible from the surface. The cleaning agent is an alkaline cleaning agent which attacks prions remaining on the surface and which attacks prions removed from the surface during the cleaning step. The invention relates to a three-dimensional scaffold material for bone tissue repair and a preparation method thereof. The three-dimensional scaffold material consists of a three-dimensional printing polymer framework with an interpenetrating macroporous structure and a protein material which serves as a filling material and has a microporous structure. The preparation method comprises the following steps: 1) preparing a polymer three-dimensional framework by a three-dimensional printer; 2) preparing an aqueous solution of protein; 3) carrying out freeze drying; and 4) adding a crosslinking agent, washing with water and then carrying out freeze drying again to obtain the product. The three-dimensional scaffold material provided by the invention is simple to operate, high in stability, free from organic solvents in the whole course and green and safe. The prepared bone tissue scaffold is good in mechanical property. The polymer in the scaffold is tightly combined with proteins. The polymer scaffold can provide an enough brute force while the protein part can provide more cell adhesion sites to benefit penetrative growth of cells, thereby promoting tissue regeneration.

The process for solubilizing proteinaceous material of the present invention includes subjecting the proteinaceous material to a sufficient amount of a basic solution to obtain a supernatant that has a basic pH and exposing the supernatant to the basic solution for a sufficient length of time and temperature for hydrolysis to occur.
