The invention relates to the technical field of fibers, and discloses a cellulose acetate composite fiber and a preparation method and application thereof in yarns, the cellulose acetate composite fiber comprises polyacrylonitrile, cellulose triacetate and cyano ester polyquaternium according to a ratio of (65-80) g: (20-35) g: (3-8) g; the compatibility between the cellulose triacetate and the polyacrylonitrile is improved by the cyano ester polyquaternium, and the breaking strength and the breaking elongation of the composite fiber are remarkably improved. The cyano ester polyquaternary ammonium salt contains a large number of antibacterial quaternary ammonium salt groups and is uniformly dispersed in matrixes of cellulose triacetate and polyacrylonitrile, so that more antibacterial sites are provided, the antibacterial property of the fiber is remarkably improved, and the antibacterial property of the fiber is remarkably improved.
