This invention belongs to the field of
plant tissue culture technology, specifically relating to a
tissue culture rapid propagation medium and method for rooting *
Carex latifolia*. The invention provides a
tissue culture rapid propagation medium for rooting *
Carex latifolia*, comprising a rooting medium; the rooting medium uses 1 / 2 MS as the
basal medium and further includes NAA 0.8 mg·L⁻¹, IBA 1.0 mg·L⁻¹,
sucrose 30 g·L⁻¹, and
agar 7 g·L⁻¹, with a pH of 5.8-6.0. The invention also provides a method for direct rooting of *
Carex latifolia* through tissue culture rapid propagation. Aseptic seedlings of *Carex latifolia* have their outer, weaker leaves removed, and the original
root system cut off, retaining 2-3 cm of the above-ground portion. These seedlings are then inoculated into the rooting medium for rooting culture to obtain rooted seedlings. After hardening off the rooted aseptic seedlings, the well-rooted *Carex latifolia* seedlings are transplanted into a substrate to form new, complete plants. The method for rapid propagation of broadleaf sedge by tissue culture provided by this invention has the characteristics of high rooting rate, large average number of roots, well-developed
root system, high transplant
survival rate, and robust regenerated seedlings.
