The fermenting medium comprises 45-80 g/L of sucrose or glucose, 23-40 g/L of soluble starch, 20-35 g/L of soybean flour or cottonseed powder, 2-4 g/L of monopotassium phosphate, 0.1-0.4 g/L of anhydrous calcium chloride, 0.1-0.3 g/L of magnesium sulfate heptahydrate, 0.01-0.037 g/L of cobalt chloride hexahydrate, 0.01-0.05 g/L of iron sulfate heptahydrate, 0.005-0.016 g/L of manganese sulfate monohydrate, and 0.005-0.014 g/L of zinc sulfate heptahydrate. Fermenting time of the liquid submerged fermentation is 55-65 h; fermenting temperature is 26-28 DEG C; dissolved oxygen control is subjected to two-stage control;dissolved oxygen value is controlled to 45-60% in the 0-24-hour bacterial growth stage of 0-24 hours, and the dissolved oxygen value is controlled to 10-15% in the 24-65-hour spore formation and maturing stage. The fermenting cycle for producing spores by liquid submerged fermentation of Purpureocillium lilacinum is shortened evidently; spore yield is increased greatly; fermentation cost of Purpureocillium lilacinum is reduced; the produced spores can well kill root-knot nematodes.
