Compared with the traditional method, the invention shortens the culture time, omits the operation procedures of filtration, and the like, and simplifies the operation steps; meanwhile, the determination data of fluorescent materials in the micropore plate can be obtained within 15s through the multifunctional microplate reader, huge samples are allowed to be simultaneously determined; and moreover, the invention has high accuracy and easy operation, stable and reliable result and good reproduction quality.

The invention discloses a method for extracting polyphenol from tobacco through a double-water-phase system. The method comprises the following steps: (1) ultrasonically extracting dry and crushed tobacco raw material, adding an inorganic salt solution and a hydrophilic micromolecule organic solvent to form the double-water-phase system, mixing, stirring, ultrasonically extracting, standing to separate the phases, wherein the polyphenol is extracted to the upper phase and the residues are distributed to the lower phase and a demarcation layer; and (2) directly sucking the organic solvent upper phase in the double water phase after ultrasonic extraction in the step (1), so as to obtain polyphenol extract; removing the inorganic salt from the polyphenol extract, and drying at a low temperature to obtain a tobacco polyphenol product. On premise of same material-liquid ratio, the one-time extraction efficiency is higher than that of a pure ethanol solution, and moreover a small amount of organic solvent is used, the organic solvent is rapid to separate, a solid residual material does not need to be filtered, the contents of protein and sugar in the extracted organic phase are greatly reduced, the obtained extraction liquid is high in concentration, small in volume, beneficial to later concentration and purification, and beneficial to industrialization.
