The method is characterized by measuring the content of the CML in food adopting high performance liquid chromatography-tandem mass spectrometry in combination with an HILIC (Hydrophilic Interaction Liquid Chromatography) column for strong-polarity CML separation under a higher organic phase proportion. Crushed food passes through an MCX (Mixed-Mode Cation-Exchange) small column for being extracted and purified after being reduced by sodium borohydride and hydrolyzed by hydrochloric acid; sample liquor is subjected to gradient elution in the HILIC column by taking methanol-water (containing 2 mM ammonium acetate and 0.1% formic acid) as a mobile phase, so that CML is separated from a sample substrate; tandem mass spectrum analysis is carried out by using an electrospray ion source, positive ion scanning and a multi-reaction monitoring mode; quantitative analysis is carried out by using D4-CML as an internal standard substance, wherein analysis time is 12 minutes, and CML has an absorption peak at the retention time of 3.47 minutes. The detecting method disclosed by the invention is high in sensitivity, high in accuracy and good in precision, does not need to carry out derivatization onto samples, simplifies sample pre-treatment, shortens detecting time and saves detecting cost.

Method for measuring interaction efficiency of quinones substances and carboxymethyl lysine in solution

The invention discloses a method for measuring the interaction efficiency of quinone substances and carboxymethyl lysine (CML) in a solution. The method comprises the following steps: respectively recording peak current and voltage characteristic values of the catechol substance solution before and after the CML is added by measuring a cyclic voltammetry curve of the catechol substance solution before and after the CML is added; and calculating the reduction rate of reduction peak current to obtain the interaction efficiency of the quinone substance in the solution and the CML. In the method,after the phenolic substance is oxidized to form the quinone substance, the quinone substance can rapidly interact with CML in a system, so that the side reaction of the quinone substance is reduced;the amount of the substance attached to the surface of an electrode is small, and the detection accuracy is high; the interference of factors such as the attachment of polyphenol and oxidation products thereof on a working electrode, the related side reaction of quinone substances and the like on the interaction of the quinone substances and CML in the prior art is reduced; and meanwhile, the method for measuring the interaction efficiency of quinone substances and carboxymethyl lysine in the solution is simple, short in analysis time, good in repeatability and convenience, low in use cost andwide in application.

Method for detecting carboxy methyl lysine (CML) in soybean sauce

The invention relates to a method for detecting carboxy methyl lysine (CML) in soybean sauce. The method comprises the following steps: firstly, carrying out detection pretreatment on CML in a soybean sauce system, namely, detection pretreatment on free-state CML and combined-state CML; carrying out detection pretreatment on the free-state CML, namely sampling, adding a methanol solution to dilute, swirling and evenly mixing, purifying through a C18 solid-phase extraction column, adding a methanol solution to elute, filtering through a micro-filtration membrane to obtain a sample 1 to be detected; carrying out the detection pretreatment on the combined-state CML, namely sampling, reducing through sodium borohydride, separating polypeptide and protein through a dialysis bag, hydrolyzing through hydrochloric acid, disacidifying through vacuum, purifying through the C18 solid-phase extraction column, filtering through the micro-filtration membrane, drying in vacuum, re-dissolving to obtain a sample 2 to be detected; and subsequently, carrying out CML detection on the sample 1 and the sample 2 to be detected respectively. A total content of the CML is sum of the content of the free-state CML and the content of the combined-state CML. The sample pretreatment method has the characteristics of elimination of interference, protection of chromatographic column, safety and reliability, and capability of completely analyzing all the CML in the soybean sauce.
