The preparation method comprises the following steps of (1) thoroughly cleaning fresh Qinghai-Tibet plateau yak skin having hair, and performing illumination through an ultraviolet lamp so as to obtain illuminated yak skin; (2) cutting the illuminated yak skin into blocks, performing extraction in boiling water,taking out the yak skin blocks after extraction, and cutting through the yak skin with a multi-stab board; (3) putting the cut-through yak skin in enzymatic hydrolysate of 0.5-10% for enzymolysis, andperforming filtration so as to obtain yak skin after enzymolysis and a first-time enzymolysis solution; (4) adding the yak skin after enzymolysis into water, and performing boiling so as to obtain filtrate; (5) adding activated carbon to the filtrate, performing sufficient stirring, then performing standing, and performing filtration so as to obtain filter residues; (6) putting the filter residues in enzymatic hydrolysate of 0.5-8% for enzymolysis, and performing filtration so as to obtain a second-time enzymolysis solution; (7) merging the first-time enzymolysis solution with the second-timeenzymolysis solution, performing boiling, and performing separation so as to obtain supernatant; and (8) filtrating the supernatant with a polyethersulfone membranes, cutting off keratin liquid of which the molecular weight is 40-75kDa and yak skin micromolecule peptide of which the molecular weight is 5kDa or below, and performing freeze drying so as to obtain yak glue. The method disclosed by the invention is simple and easy to implement.
