The method comprises the following steps: dripping a modification liquid to the surface of a glassy carbon electrode, baking, inserting the glassy carbon electrode serving as a working electrode, an auxiliary electrode and a reference electrode into a mixed solution of glycine and sodium nitrate, and repeatedly scanning, so that the surface of the glassy carbon electrode is coated with a polyglycine-multiwalled carbon nanotube / naphthol / epoxy phosphate film; inserting a modified electrode serving as a working electrode into a base solution containing lead ions, giving a voltage-current curve summary chart by virtue of an electrochemical workstation, and reading the current peak of the concentration of each lead ion; and drawing a relational graph between the lead ion concentration and the current peak, obtaining a linear relation formula between the lead ion concentration and the current peak, igniting circular filter paper of a PM2.5 sample to be burnt into ash, dissolving the ash in an HNO3 base solution, inserting the modified electrode serving as a working electrode into a to-be-detected base solution, giving a voltage-current curve chart by virtue of the electrochemical workstation, reading the current peak of the lead ions, and calculating the lead ion concentration through a formula.
