In biology, quorum sensing is the ability to detect and to respond to cell population density by gene regulation. As one example, quorum sensing (QS) enables bacteria to restrict the expression of specific genes to the high cell densities at which the resulting phenotypes will be most beneficial. Many species of bacteria use quorum sensing to coordinate gene expression according to the density of their local population. In similar fashion, some social insects use quorum sensing to determine where to nest.

Isolated nucleotide molecule and method of sensing and killing of pathogenic microorganism

The present invention relates to an isolated nucleic acid molecule comprising (a) a first nucleotide sequence encoding a protein that detects the presence, amount or both of a pathogenic microorganism by forming a complex with a quorum sensing molecule produced by said pathogenic microorganism, (b) one or more second nucleotide sequence said one or more second nucleotide sequence being under control of a promoter that is induced by the complex of the protein encoded by the first nucleotide sequence and the quorum sensing molecule produced by said pathogenic microorganism and encoding (i) an antimicrobial peptide, wherein the antimicrobial peptide is effective against the pathogenic microorganism detected by the protein encoded by the first nucleotide sequence; and/or an antibiofilm enzyme wherein the antibiofilm enzyme is effective against the pathogenic microorganism detected by the protein encoded by the first nucleotide sequence; and (c) optionally a third nucleotide sequence encoding a protein that controls the motility of the host organism, wherein the protein that controls the motility of the host organism directs the motility of the host organism towards said pathogenic microorganism. A recombinant microorganism comprising the isolated nucleic acid molecule and a method of sensing and killing pathogenic microorganisms is also described.

The invention belongs to the field of hygiene and relates to a bacillus subtillus degrading bacterial colony sensing signal and use of bacillus subtillus degrading bacterial colony as an antiseptic, in particular to bacillus subtillus degrading gram-negative bacterial colony sensing signal and the formation of a bacterial biofilm of the bacillus subtillus degrading gram-negative bacterial colony. In the use of the degrading bacterial colony sensing signal, a proper concentration of the bacillus subtillus is 5*102 to 5*104cfu / ml. The bacillus subtillus is further used as antiseptic to inhibit the bacteria such as pseudomonas, agrobacterium, enterobacteriaceae and vibrionaceae. The antiseptic can be ointment for external use, liquid preparation for external use, oral tablet, oral liquid, disinfectant, clearing agents or water quality improving agent. The consistency of the bacillus subtillus serving as the antiseptic is 105 to 106 cfu / g. The use of the bacillus subtillus provided by the invention and the antiseptic inhibit the formation of biofilm of disease-causing bacteria and the growth of bacteria by using microbial quorum-sensing inhibition instead of preventing and sterilizing bacteria by compound which is adopted by most of the conventional antiseptic; therefore, the problem of drug resistance in the antimicrobial process is avoided.

The invention relates to the field of analytical chemistry, and discloses a method for simultaneously determining bacterial quorum sensing AHLs molecules. A to-be-detected sample is added in a high performance liquid chromatography-tandem mass spectrometer for gradient elution. The elution conditions are that the elution is carried out by using an A phase (methanol containing 10 mmoI/L ammonium acetate and 0.2 % formic acid) and a B phase (a water solution containing 10 mmoI/L ammonium acetate and 0.2 % formic acid); parameters for the gradient elution are as follows: at 0 min, the content of the A phase is 20% of and the content of the B phase is 80% and the content reaches a preset proportion at the moment; from 0 min to 8 min, two flowing phases are changed into 100% of the A phase and 0% of the B-phase at an average rate; from 8 min to 10.1 min, the content of the A phase is 100% of and the content of the B phase is 0%; at 10.1 min, the content of the A phase is adjusted to 20% of and the content of the B phase adjusted to 80% instantly; and elution is continuously performed to 15 min. The method has good linear relationship, a detection unit of 0.1-1.0 ng/L, has high accuracy and sensitivity, and simple operations.
