The method comprises the steps as follows: under aerobic and dark conditions, pycnoporus sanguineus CGMCC No. 2932 is cultivated in a sterile fermentation medium containing a carbon source, a nitrogen source and inorganic components, the fermentation medium comprises cornmeal, bran powder, ammonium tartrate, monopotassium phosphate, magnesium sulfate heptahydrate, sodium succinate, copper sulfate pentahydrate, potassium chloride hexahydrate, zinc vitriol, a surfactant, an antifoaming agent and water, the ventilation quantity is adjusted in a fermentation process, and nutrients are fed-batched. The method for preparing the laccase-containing fermentation broth by pycnoporus sanguineus solves the problems of poor reproducibility, low enzyme activity and the like in the prior art, and has the advantages of good reproducibility, high activity of produced laccase, purer produced enzyme system, good enzyme temperature stability, high pH adaptability and the like; and the laccase can be widely applied to the fields of dye degradation, biobleaching, wood gluing, ecological restoration and the like.
