The invention belongs to the technical field of preparation of drugs, and specifically relates to quercetin-modified nano sulfur as well as a preparation method and application thereof to an anti-AD (Alzheimer's Disease) drug. According to the preparation method disclosed by the invention, quercetin is modified on nano sulfur by utilizing the microsize and a high surface energy effect of nano sulfur, not only a function of the quercetin for reducing endoplasmic reticulum stress in cells of the quercetin is utilized, but also the water solubility is improved, so that quercetin-modified nano sulfur integrating high biocompatibility and an AD treatment effect is obtained. The invention provides the quercetin nano sustained-release preparation which is prepared by a spontaneous polymerization method by taking acetic acid as an initiator and is safe and effective in resisting silica-dust-caused pneumoconiosis fibrosis. According to the method, no toxic organic solvent is used for dissolving water-insoluble quercetin, instead, glacial acetic acid is used for dissolving the quercetin. The glacial acetic acid is weak acid, the acidity value changes slowly during pH adjustment, the pH value is easy to control, while strong acid hydrochloric acid cannot be used, acetic acid is easy to ionize acetate anions in water, the anions can be used as an initiator, and when a monomer is added into water, the monomer is automatically initiated to be polymerized into a polymer and nanoparticles can wrap quercetin, so that the solubility and bioavailability of the quercetin are improved.
