Gelatin/Elastin Composites for Peripheral Nerve Repair

Neurosupportive materials that possess strong tissue adhesion were synthesized by photocrosslinking two polymers, gelatin methacryloyl (GelMA) and methacryloyl-substituted tropoelastin (MeTro). The engineered materials exhibited tunable mechanical properties by varying the GelMA / MeTro ratio. In addition, GelMA / MeTro hydrogels exhibited 15-fold higher adhesive strength to nerve tissue ex vivo compared to traditionally used fibrin-based materials. Furthermore, the composites were shown to support Schwann cell (SC) viability and proliferation, as well as neurite extension and glial cell participation in vitro, which are essential cellular components for nerve regeneration. Finally, subcutaneously implanted GelMA / MeTro hydrogels exhibited slower degradation in vivo compared with pure GelMA, indicating its potential to support the growth of slowly regenerating nerves. Thus, GelMA / MeTro composites may be used as clinically relevant biomaterials to regenerate nerves and reduce the need for microsurgical suturing during nerve reconstruction.

The invention belongs to the field of biomedical engineering, and relates to a tissue engineered artificial optic nerve conduit and a preparation method thereof. The tissue engineered artificial optic nerve conduit comprises a hollow conduit woven from polylactic-polyglycolic acid filaments (PLGA), polyglycolic acid (PGA) filaments and transgenic schwann cell suspension, wherein chitosan polymer is coated on the surface of the conduit; an external bracket of artificial optic nerve is constructed by the PLGA hollow conduit; and the PGA filaments are filled in an internal bracket in which the artificial optic nerve is constructed. The conduit can effectively promote regeneration of damaged optic nerve axon, and has the advantages of persistently providing nerve nutritional support, guiding the regenerated nerve axon and the like; the conduit has high tissue compatibility and safety; the specifications of the transfected gene and the conduit can be adjusted according to requirements; and the conduit and the method can be used for experimental research of optic nerve regeneration and expected to promote regeneration and restoration of damaged optic nerve in clinical use.
