The present invention provides a method for treating osteoporosis in a subject in need of such treatment, including administering orally to the subject an effective amount of an extract product of a Dioscorea species capable of promoting proliferation and differentiation of bone marrow cells in the subject, wherein the Dioscorea species is characterized by a randomly amplified polymorphic DNA (RAPD) fingerprint including 14 DNA bands ranging from 428 bp, 452 bp, 537 bp, 602 bp, 723 bp, 817 bp, 934 bp, 1140 bp, 1242 bp, 1478 bp, 1641 bp, 1904 bp, 2151 bp and 2918 bp, respectively, when genomic DNA of the Dioscorea species is amplified with a primer of SEQ ID NO: 9, and the extract product is prepared by a process including the steps of: (a) extracting a tuber of the Dioscorea species using an alcohol-based solvent in the presence of acetic acid; (b) subjecting the resultant product obtained in step (a) to a separating treatment to obtain a soluble fraction; and (c) removing solvent from the soluble fraction obtained in step (b) extract fractions of Dioscorea sp.

The strain liquid is fermented by a common culture medium for 48 to 60 hours, the chloroform extract of the fermentation liquor is the diosgenin, and the diosgenin has the activity of inhibiting pathogenic bacteria and resisting tumor cells. The method comprises the following steps: putting dried dioscorea powder into a reaction kettle, adding a ternary two-phase solution consisting of an aluminum trichloride aqueous solution, ethanol and petroleum ether, carrying out homogenizing, carrying out reacting for 0.5-3.0 hours at 130-180 DEG C, filtering the reaction solution, filtering out residues, separating out a petroleum ether layer, carrying out rotary evaporation, and carrying out recrystallizing to obtain the diosgenin. The invention belongs to the technical field of a microorganism secondary metabolite, and discloses a method for preparing diosgenine by a fermenting dioscorea zingiberensis endophyte. Diosgenine is obtained and separated from fermentation liquor of a dioscorea zingiberensis endophyte. The method comprises the following steps: 1) inoculating a strain culture medium with the dioscorea zingiberensis endophyte for strain culture to obtain a fermentation strain; 2) inoculating a fermentation tank containing a fermentation culture medium with the fermentation strain, starting stirring, and setting fermentation conditions of the fermentation temperature being 24-32 DEG C, the fermentation pH being 5.5-8.5, and the ventilation amount being 1.0-5.5 L / min; and 3) performing acidolysis to the obtained fermentation liquor, and carrying out neutralization to obtain diosgenine. The method for preparing the diosgenin comprises the following steps of culturing fusarium fungus by using a liquid culture medium containing dioscorea zingiberensis to obtain the diosgenin, wherein the fusarium fungus is a fusarium strain (Fusarium sp.) CPCC 400226. Experiments provethat the yield of the diosgenin is 2.48 + / -0.21% after the fusarium sp. CPCC 400226 is cultured for 11 days at 28 DEG C by adopting the liquid fermentation culture medium disclosed by the invention ina liquid fermentation manner. The method can be used for clean production of diosgenin.
