This invention provides a method for culturing aerobic methanogenic microorganisms in
mangrove sediments, belonging to the field of microbial technology. The method comprises five steps: culture medium preparation, sample pretreatment, mud
system preparation, aerobic environment construction, and isothermal incubation. The culture medium contains basic salts, macroelements, microelements, and vitamins adapted to the
brackish water environment of mangroves. By constructing a stable aerobic culture
system, the efficient enrichment of aerobic methanogenic microorganisms in
mangrove sediments is achieved. This invention is specifically adapted to the environmental characteristics of
mangrove sediments, filling the gap in aerobic methanogenic
microorganism enrichment and cultivation technology in mangrove ecosystems. It can stably achieve the cultivation and enrichment of aerobic methanogenic microorganisms, providing reliable
technical support for elucidating the aerobic
methanogenesis process in coastal mangrove wetlands, isolating and purifying aerobic methanogenic functional microorganisms, and expanding the methanogenic microbial resources of mangrove ecosystems.
