A preparation method of high purity capsaicin monomer comprises the following steps: using alcohol to extract capsaicin from pepper powder or pepper essential oil, then using macroporous resin to enrich the extract so as to obtain pepper total alkaloid; adopting a high speed counter-current chromatography method to separate the pepper total alkaloid, wherein the separating solvent system is composed of alkane, ester, alcohol, and water, and acid and alkali can be added too; adding the solvent system into a separating funnel, fully vibrating the separating funnel, allowing the separating funnel to stand still so as to divide the solution to different layers, collecting the upper-layer solvent as the stationary phase and the lower-layer solvent as the mobile phase; dissolving a sample in a mixed solvent of the upper-layer solvent and the lower-layer solvent, injecting the sample into a detector, separating, and collecting the target components according to the spectrogram on the detector so as to obtain the capsaicin monomer and dihydrocapsaicin monomer. The invention discloses a method of separating and purifying capsaicin and dihydrocapsaicin from pepper fruits. The method comprises the following steps: performing supercritical carbon dioxide extraction of pepper fruits; performing reduced-pressure concentration to obtain capsicum oleoresin; performing methanol solution extraction and alkali extraction and acid deposition of the capsicum oleoresin to obtain a capsaicin substance crystal rich in capsaicin and dihydrocapsaicin; with n-hexane-ethyl acetate-methanol-water as a solvent system and high-speed counter-current chromatography (HSCCC)for preparation, monitoring the purity by high performance liquid chromatography; according to the HSCCC as well as the eluent with relatively high purity, performing reduced-pressure concentration and vacuum drying to obtain capsaicin with purity higher than 97.0% and dihydrocapsaicin with purity higher than 96.0%, wherein the extraction efficiencies of the capsaicin and the dihydrocapsaicin areboth higher than 93%, and the recovery rates are both higher than 70%. The method disclosed by the invention makes full use of the pepper seeds, has the advantages of easiness in operation, low cost,high repeatability, high efficiency and high product purity, and can be applied to large-scale production.

The method is obvious in effect, simple in process and easy and convenient to operate for separating capsaicin and dihydrocapsaicin, the solvent and resin can be recycled, no emission of industrial wastewater, waste gases and residues is generated, and the method is suitable for large-scale industrialized production.

Method for preparing high-purity capsaicin monomer

The invention discloses a method to prepare high-purity capsaicin monomers. The separation solvent system is composed of alkyl halies, alcohol, water, etc. The separation solvent system is arranged inthe separating funnel, and is shaken and then kept still for stratification, the upper phase is taken as fixed phase and the lower phase as mobile phase. A proper dosage of sample is weighed and dissolved in the mixed solution of the upper phase and the lower phase. Firstly, the fixed phase is pumped to fill fully a chromatographic separation column at the specified flow rate, and then the head end of the column is connected with a six-way sample-feeding valve. The speed controller is started; when the rotational speed reaches to 800r / min, the sample solution is pumped into the mobile phase atthe flow rate of 2.0mL / min; when the mobile phase effuses from the chromatographic column, the sample is loaded, and each separated peak is collected; high-performance liquid chromatography (HPLC) is adopted to measure the purity, and three monomers i.e. dihydrocapsaicin, capsaicin and nordihydro-capsaicin are achieved. The method is applicable to the preparation of such three high-purity capsaic in monomers by the crude extracts of the capsaicin obtained through various approaches. The method is characterized in simple operation, low production cost and free of organic solvent residue; and the method can be promoted for the preparation of high-purity capsaicin monomers.
