The invention discloses a colibacillus O157:H7
DNA label-free detection device and method based on LAPS and ZnO
nanorod array. The device comprises a Faraday shield box, a loading table, a
semiconductor laser, an LAPS sensor, a manostat, a lock-in
amplifier, an acquisition card, and a computer. The method comprises of performing bacterial culture, extracting colibacillus O157:H7
DNA, manufacturing aLAPS sensor and modifying a ZnO
nanorod; modifying probe chain
DNA on a surface of the LAPS sensor, and scanning an I-
V curve; fitting an optimal
calibration curve detecting concentration of O157:H7DNA through traversing and a least square method; and analyzing an I-
V curve of a to-be-tested sample solution and
voltage offset V and substituting into the
calibration curve, and calculating concentration of the to-be-tested sample. The method realizes
label-free detection of O157:H7 DNA, and has advantages of short time, low cost, convenience in operation, 3D site for O157:H7 DNA
hybrid, and high sensitivity.
