In the present invention, hydrogen peroxide is first used to destroy the hydrogen bond between the konjac polysaccharide molecules, then cellulase is used to degrade it, and 60% ethanol alcohol precipitation is separated and purified to prepare the konjac polysaccharide degradation product KGM-with uniform molecular weight 1k and KGM‑5k components. The effects of konjac polysaccharide degradation products KGM-1k and KGM-5k on the survival of probiotics were investigated for the first time under simulated gastric juice, small intestinal juice, vacuum freeze-drying and storage conditions at different storage temperatures (4℃ and 25℃). The results showed that KGM-1k and KGM-5k, the degradation products of konjac polysaccharide, could enhance the stress resistance of probiotics, improve the number of viable bacteria and the survival rate in the digestive tract of probiotics under vacuum freeze-drying and storage conditions at different storage temperatures. Furthermore, it can tolerate gastrointestinal juices entering the predetermined part of the intestine to achieve colonization, which is of great significance for improving the biological efficacy of probiotics.
