A homeostatic system may be used for production of ATP, where the required high energy phosphate bonds are generated in situ, e.g. through coupling with an oxidation reaction. The homeostatic energy source will typically lack high energy phosphate bonds itself, and will therefore utilize free phosphate in the reaction mix during generation of ATP. A Yersinia pestis virulence regulator TyrR and applications thereof are provided. It is found that virulence of TyrR-deleted Yersinia pestis strains is lowered about 10000 times than virulence of wild strains, lack of the TyrR has no influence on in-vitro growth of the Yersinia pestis, and the TyrR can achieve negative regulation of transcription of five aromatic amino acid metabolism genes comprising aroF-tyrA, aroP, aroL and tyrP, and achieve positive regulation of two acid response genes which are hdeB and hdeD, so that the TyrR is proved to be a Yersinia pestis virulence regulator. In an environment having a pH value of 5.0, the survival rate and a growth speed of a Yersinia pestis TyrR mutant strain are lower than that of a wild strain, thus proving that the acid resistance of a TyrR mutant strain is lowered and that the TyrR regulator is closely related to acid resistance of the Yersinia pestis. Applications of the Yersinia pestis virulence regulator TyrR in preparation of anti-plague vaccines, and applications of the virulence regulator TyrR in preparation of medicines adopting the TyrR as a target are provided.

It is shown by dimensional electrophoresis results that when a long term low nitrogen treated group is compared with the reference group, 7 differential protein spots are detected, one up-regulation is detected, and 6 down-regulation is detected; when a long term high nitrogen treated group is compared with the reference group, 6 differential protein spots are detected, one up-regulation is detected, and 5 down-regulation is detected. Mass spectrum identification and function classification are adopted, it is found that the proteins are a protein and amino acid metabolism associated protein, an energy metabolism associated protein, an expression regulation and control and defense and stress resistance associated protein, a cell metabolism associated protein, and a protein with unknown functions.

Bacterial Metastructure and Methods of Use

Although metabolic networks have been reconstructed on a genome-scale, the corresponding reconstruction and integration of governing transcriptional regulatory networks has not been fully achieved. Here such an integrated network was constructed for amino acid metabolism in Escherichia coli. Analysis of ChlP-chip and gene expression data for the transcription factors ArgR, Lrp, and TrpR showed that 19 / 20 amino acid biosynthetic pathways are either directly or indirectly controlled by these regulators. Classifying the regulated genes into three functional categories of transport, biosynthesis, and metabolism leads to elucidation of regulatory motifs constituting the integrated network's basic building blocks. The regulatory logic of these motifs was determined based on the relationships between transcription factor binding and changes in transcript levels in response to exogenous amino acids. Remarkably, the resulting logic shows how amino acids are differentiated as signaling and nutrient molecules. This reveals the overarching regulatory principles of the amino acid stimulon.
