The invention discloses a screening method of grape wine yeast with low-yield ethyl carbamate and application of grape wine yeast in the production of grape wine. The screening method comprises the following steps of: screening out a bacterial strain with the low-yield ethyl carbamate from active dried yeast and mutant strains by adopting a structural analogue of arginine, i.e., canavanine resistance flat plate, wherein the activity dried yeast can be separated from nature or can be purchased in market, and the mutant strains are obtained through the mutagenesis of chemical mutagen. The method is designed by a yeast amino acid metabolism theory and is simple and convenient; and a target bacterial strain can be obtained through rapid separation. According to the screening method, the obtained resistance bacterial strain B1 can be numbered as F15-1-15 bacterial strain (CGMCC (China General Microbiological Culture Collection Center) NO. 4727) and is applied to the brewing of the grape wine in a small-sized fermented container; and on the basis of excellent brewing performance of the grape wine, the bacterial strain has the advantages of being capable of effectively and obviously reducing the content of the ethyl carbamate in the production of the grape wine and having the potential of industrial production and application.

The invention discloses a method for analyzing the influence of taurine on the growth of tilapia on the basis of NMR metabonomics technology. The method comprises: collecting fish muscle samples and collecting the one-dimensional nuclear magnetic resonance hydrogen spectrum of each sample to obtain the original spectrogram of each sample; performing data preprocessing on each original spectrogramto obtain a two-dimensional data matrix; performing multivariate statistical analysis on the two-dimensional data matrix to obtain small molecular metabolites of each tilapia muscle tissue under the influence of different taurine addition amounts; and obtaining a metabolite change law through the identification and pattern recognition analysis of the metabolites, and screening biomarkers related to sugar metabolism, lipid metabolism and amino acid metabolism. According to the method for analyzing the influence of the taurine on the growth of the tilapia on the basis of the NMR metabonomics technology, the nuclear magnetic resonance metabonomics technology is used for analyzing the influence of the taurine on the growth of the tilapia, so that the metabolic difference of tilapia muscles under the influence of different taurine addition amounts can be rapidly, accurately, and comprehensively obtained, and the addition accuracy of taurine feed in tilapia breeding and the influence on thegrowth of the tilapia can be effectively evaluated.

The invention discloses a method used for measuring influences of long term applications of high and low nitrogen fertilizers on corn kernel albumin. According to the method, proteomic method is adopted to study long term positioned fertilization of high and low nitrogen fertilizers on summer corn Luyu 16 kernel albumin. It is shown by results that: the albumin content of a reference group is higher than those of two processed groups, and obvious difference strips are observed in quantificative unidirectional SDS-PAGE. It is shown by dimensional electrophoresis results that when a long term low nitrogen treated group is compared with the reference group, 7 differential protein spots are detected, one up-regulation is detected, and 6 down-regulation is detected; when a long term high nitrogen treated group is compared with the reference group, 6 differential protein spots are detected, one up-regulation is detected, and 5 down-regulation is detected. Mass spectrum identification and function classification are adopted, it is found that the proteins are a protein and amino acid metabolism associated protein, an energy metabolism associated protein, an expression regulation and control and defense and stress resistance associated protein, a cell metabolism associated protein, and a protein with unknown functions.

Amino acid metabolome in each of tissues of the brains of para-chlorophenylalanine (PCPA) insomnic model rats before and after gavage with the sugemule is analyzed by applying a gas chromatography-mass spectrometer (GC-MS) method; endogenous compounds are identified by astandard fingerprint bank; changes of metabolite maps of all parts of the brains are analyzed and related metabolic markers are assayed by employing principal component analysis (PCA) and partial least squares-discrimination analysis (PLS-DA) modes; a targeting metabolomics research is conducted by applying liquid chromatograph-mass spectrometer (LC-MS) to explore content changes of neurotransmitters including 5-hydroxytryptamine, gamma-aminobutyric acid (GABA), glutamic acid (Glu) and the like in the brain tissues and related metabolites; by seeking for difference metabolites related with thecurative effect of the sugemule and metabolic pathways of the difference metabolites from brain tissue samples of clinical rats, the anti-insomnia action mechanism and an action site of the sugemuleare specified; and a metabolomics research means is applied for the first time by the amino acid metabolomics-based method to preliminarily confirm the action mechanism of the sugemule regulating andcontrolling a sleep-wake rhythm by a neurotransmitter system.

Method for screening microorganisms by using supermolecular fluorescence microfluidic technology

The invention provides a method for detecting the concentration of amino acid generated in the metabolic process of microorganisms based on a supramolecular fluorescence system and realizing high-throughput screening of the microorganisms by combining a droplet microfluidic technology. The method comprises the following steps of: (1) generating water-in-oil droplets on a micro-fluidic chip, and researching the influence of amino acid on the fluorescence intensity of a cucurbituril/fluorescent dye supramolecular system in the micro-fluidic droplets; (2) wrapping and culturing microorganisms byutilizing a droplet microfluidic technology, then injecting cucurbituril/fluorescent dye into microfluidic droplets, and distinguishing the microorganisms with different amino acid metabolic capabilities according to the change of the fluorescence intensity of the droplets; and (3) carrying out high-throughput screening on the microorganisms with different amino acid metabolic capabilities by utilizing a microfluidic droplet screening technology according to the difference of fluorescence intensity of each microfluidic droplet. Fluorescence of the supramolecular system is researched in the microfluidic droplets for the first time and applied to detection of the metabolite concentration of the microorganisms, and high-throughput screening of the microorganisms with different metabolic capabilities is established accordingly. The detection method has the advantages of high sensitivity, high speed and the like, with the screening flux reaching millions of droplets per hour. By adopting the high-throughput screening method disclosed by the invention, microorganisms capable of producing target products at high yield are obtained.

Application of sodium dichloroacetate in preparation of low protein feed for pigs

The invention discloses application of sodium dichloroacetate in preparation of a low protein feed for pigs. The inventor firstly classifies raw materials into three types; the first type is known as a micro-class raw material with the mixing proportion less than 2 percent, the second type is known as a medium-class raw material with the mixing proportion greater than 2 percent and less than 5 percent, and the third type is known as a large-class raw material with the mixing proportion greater than 5 percent. The low protein feed for the pigs is prepared by the following steps: firstly, mixing the first type of feed raw materials, fully and uniformly mixing the raw materials through a stirrer, then mixing the raw materials with the medium-class raw materials, thus obtaining a mixture, and finally, mixing the mixture with the large-class raw materials, thus obtaining the amino acid metabolism thrift-based low protein feed for the pigs; granulating the mixture according to the particle size of 8 to 12 mm; and transferring the particles into a drying machine for drying at the temperature of 95 to 105 DEG C, thus preparing a particulate feed finished product. The feed is wide in source, low in cost and comprehensive in nutrition; the level of coarse proteins in the feed can be obviously reduced; the growing performance of the animal cannot be affected; and the low protein feed is applicable to large-scale production.
