The method is realized by preparing culture materials by using basic culture materials and a selenium-enriched culture material additive. The method is characterized in that the selenium-enriched culture material additive accounts for 1-10% of the total weight of dry culture materials; the selenium content is 1-20mg/kg; the sizes of grains in the selenium-enriched culture material additive are in multi-size regional distribution in the range from 1.0mm to 5.0mm; and the selenium-enriched culture material additive is prepared from the straws of such crops as selenium-enriched wheat or corn with selenium content being 1-80mg/kg by grinding. The method has the following advantages: the agarics cultured by the method have high selenium absorption and utilization efficiency; the highest content of selenium in the fruiting bodies of the agarics can be 20 times that of selenium in the fruiting bodies of common culture groups; the organic selenium is as high as 90% of the total selenium; and in the growth periods of the fruiting bodies, the relative standard deviation (RSD) of the selenium contents of the fruiting bodies can be strictly controlled within 20%.
