The invention relates to a method for artificially inducing resistance to jujube witches
broom, and the method comprises the following steps: longitudinally
cutting a
cut at the lower end of the stem section of a cultured
plantlet in a healthy group of a susceptible variety, wherein the stem section is utilized as a scion; removing leaves and axillary
bus from the stem section of the cultured
plantlet in a group infected with the jujube witches
broom, and
cutting one surface of the upper end of the stem section into a
diagonal plane, wherein the stem section is utilized as a
rootstock; then inserting the scion, aligning the
rootstock with a scion formation layer, and using an
aluminum foil to bind a
grafting port, thus acquiring a micro-grafted
plantlet; inserting into an MS (Murashige and Skoog) culture medium without any
plant growth regulator for culture; adding 1mg / ml of
tetracycline to kill pathogens in the micro-grafted plantlet after
graft survival and afference of the pathogens into the scion,
cutting the scion, transferring into the MS culture medium which is additionally added with 1.0mg / mL of BA (6-benzyl adenine) and 0.5mg / mL of IBA (indole-3-
butyric acid) for rejuvenation, and further
grafting the scion after the rejuvenation onto the
rootstock of the cultured plantlet in the group with the jujube witches
broom; and continuously performing the induction containing micro-
grafting inoculation,
processing with the
tetracycline, rejuvenation and micro-grafting
inoculation above three times, thus obtaining a
plant with resistance to the jujube witches broom. By utilizing the method, the
plant with the resistance to the jujube witches broom can be rapidly induced and obtained. By adopting the method, the resistance to the jujube witches broom of jujube witches broom-sensitive good jujube tree varieties can be rapidly improved in an orientation way, thus the method provided by the invention has very broad application prospects.
