Preparation method and process for antibacterial modification of ceramic whiskers

The invention relates to a preparation method and process for antibacterial modification of ceramic whiskers and in particular provides a preparation method for fusion-coating of zinc oxide nanoparticles having different patterns on the surface of hydroxyapatite whiskers. The preparation method is characterized by adding dropwise an ethanol solution of a zinc salt into a beaker in which an ethanol solution of hydroxyapatite whiskers is held, performing stirring, rotating, ultrasonic treatment, drying and calcination procedures to make zinc oxide-hydroxyapatite whiskers. The ceramic whiskers have antibacterial effects. The zinc oxide nanoparticles have good antibacterial effects and the antibacterial rate against Streptococcus mutans reaches (93.58 plus or minus 5.95) percent; and the hydroxyapatite whiskers have high strength. After modification, zinc oxide nanoparticles, nanospheres, nanorods, microspheres and diamond-shape units are uniformly distributed on the surface of the hydroxyapatite whiskers by fusion coating, the particle size is 30 nm to 400 nm, the surface area and the roughness are greatly improved, and the mechanical embedment force between the zinc oxide nanoparticle and the matrix is increased. Therefore, the zinc oxide-modified hydroxyapatite whisker can serve as an ideal reinforcing material with antibacterial effect.

The invention discloses a probiotics toothpaste composition capable of improving the health of oral cavity. The composition comprises a first toothpaste component and a second toothpaste component; wherein the first toothpaste component comprises probiotics and mannan oligosaccharide, and the second toothpaste component comprises enzymes, mannatide, and a diester type surfactant; and the first toothpaste component and the second toothpaste component are individually packed, and the first toothpaste component and the second toothpaste component are mixed and blended when being used on the teeth. Mannan oligosaccharide and degradation product of mannatide can provide carbon sources for the growth of probiotics, the pathogenic bacteria in oral cavity cannot utilize mannan oligosaccharide, moreover, mannan oligosaccharide can absorb pathogenic bacteria so as to starve the pathogenic bacteria; at the same time, alkaline polypeptide in degradation product of mannatide can cut off glycan chain of cell walls of bacteria; and the diester type surfactant has a strong cleaning and stain-removing performance, and is capable of effectively remove the residual bacterium bodies. After application, the number of pathogenic bacteria especially streptococcus mutans is greatly reduced, and the health of oral cavity is prominently improved.

The production process comprises the following steps: preparation of a solution I: heating purified water, then adding sodium benzoate, cetylpyridinium chloride, sodium fluoride, sucralose, noncrystalline sorbitol, nisin and brilliant blue and carrying out stirring for 20 to 25 min; preparation of a solution II: heating hydrogenated castor oil RH40 and propylene glycol to a fusion state, adding peppermint essence and carrying out uniform mixing under stirring so as to obtain the solution II; preparation of a solution III: uniformly mixing PEG-400, xanthan gum and sodium alginate in a ratio prescribed in a formula under stirring so as to obtain the solution III; and preparation of the probiotic periodontal care gel: adding the solution II into the solution I, carrying out uniform mixing under stirring, then adding the solution III, carrying out stirring for 25 to 30 min so as to form a semi-finished product, taking a sample for laboratory test so as to obtain a semi-finished cetylpyridinium chloride product, carrying out canning when the semi-finished cetylpyridinium chloride product is determined to be qualified and carrying out warehousing after a finished product is determined to be qualified, wherein if cetylpyridinium chloride is insufficient, cetylpyridinium chloride is added, a sample is taken for detection until the semi-finished product is qualified and then canning is carried out, and if the finished product is determined to be unqualified, the finished product is discarded. Lactobacillus can reinforce inhibitory effect of sodium fluoride on Streptococcus mutans, reduce accumulation of dental plaques, prevent gingivitis and carious teeth and improve the balance of microfloras in the oral cavity.

The invention discloses a preparation method and application of a quinoxaline amide compound used as a streptococcus mutans biological membrane inhibitor and relates to the technical field of bacteriostatic drug screening. The inhibitor consists of compounds 2-(4-methoxy-phenyl)-N-(3-{[2-(4-methoxy-phenyl) ethyl] imido}-1,4-dihydro-2-quinoxaline subunit) ethylamine and dimethyl sulfoxide (DMSO). The preparation method comprises the following steps: (1) dissolving 50mg of powdery 2-(4-methoxy-phenyl)-N-(3-{[2-(4-methoxy-phenyl) ethyl] imido}-1,4-dihydro-2-quinoxaline subunit) ethylamine in 1mL of a dimethyl sulfoxide (DMSO) solvent to prepare an inhibitor mother solution of which the initial concentration is 50mg/mL; (2) performing gradient dilution on the inhibitor mother solution by using dimethyl sulfoxide (DMSO), and adding into a brain heart infusion (BHI) liquid culture medium to make sure that the final concentrations of the inhibitor are respectively 0.05mu g/mL, 0.5mu g/mL, 5mu g/mL and 50mu g/mL and the final concentrations of the solvent DMSO are all 1% (v/v). Application tests show that the inhibitor can be used as an oral mouth streptococcus mutans biological membrane inhibitor and a candidate targeted drug for a novel anti-caries strategy.

By adopting the CRISPR-B gene editing method disclosed by the invention, the synthesis of streptococcus mutans extracellular polysaccharides and the formation of a biological film can be inhibited to the greatest extent.
