The invention discloses an
exosome freeze-
drying protective agent and application of the
exosome freeze-
drying protective agent in an oral
soluble film. The protective agent is prepared from the following
solid matters in parts by weight: 1 to 20 parts of zwitterionic
polymer containing
choline phosphate side groups, 10 to 80 parts of polyoxazoline
polymer and 1 to 20 parts of volatile
ammonium salt. The zwitterionic
polymer containing the
choline phosphate side group forms zwitterionic pair interaction through charge arrangement
complementation between a
choline phosphate group and a
phosphatidylcholine head group on the surface of an
exosome membrane, so that directional protection is provided for the exosome
membrane structure; after the polyoxazoline polymer is freeze-dried, a glassy state curing matrix is formed to embed and support the exosome; the volatile
ammonium salt is decomposed and volatilized in the freeze-
drying secondary drying stage, and a communicated pore channel is formed in the membrane body. The invention also provides an application of the protective agent in preparation of an exosome freeze-drying preparation or an exosome oral
soluble film, and the obtained oral
soluble film can be rapidly disintegrated, can maintain the
structural stability of the exosome, and is suitable for normal-temperature storage and rapid administration.
