A culture medium comprises 10 to 90 weight percent of debarked magnolia officinalis branch wood pellets, the total C/N ratio of the culture medium is (50-80):1, and the pH value is 5 to 8. The method comprises the following steps of: uniformly mixing the magnolia officinalis branch wood pellets which are fully soaked with water and other components of the culture medium, sterilizing and inoculating; and culturing under environmental conditions adapting to the growth habit of the lucid ganoderma or auricularia polytricha strains so as to obtain the finished product of the lucid ganoderma or auricularia polytricha. The method solves the problem of selection of culture media during conventional cultivation of the lucid ganoderma or auricularia polytricha, also solves the problem of recycling massive debarked magnolia officinalis branches which are used as medicines, and changes waste materials into valuable things; meanwhile, the conventional cultivating and operating method is not needed to be changed, the yield of the lucid ganoderma or auricularia polytricha can be improved, and the method has high social significance and commercial value.
