The invention relates to a method for detecting the content of free
sodium hyaluronate in medical cross-linking
sodium hyaluronate gel. The detection method comprises the following steps: weighing a certain number of samples, adding an appropriate quantity of a PBS solution, enabling the weighed samples to be dissolved in the PBS solution so as to obtain a solution, uniformly mixing the solution, filtering the
mixed solution with a
filtration membrane, rinsing a vials with an appropriate quantity of PBS, taking a filtrate, and determining the content of glucuronic acid; (2) preparing a series of glucuronic acid standard fluid; (3) separately adding 5 mL of a 0.025 mol / L
sodium tetraborate
sulfuric acid solution to a tube loaded with the series of the standard fluid and a sample tube, performing uniform mixing, performing heating in boiling water bath for 10 minutes, performing cooling to
room temperature, adding 0.2 ml of a
carbazole ethanol solution to the tubes, performing uniform mixing, performing heating in boiling water bath for 15 minutes, performing cooling to
room temperature, and determining the
absorbance of the standard tube and the sample tube at the position of 530 nm with a spectrophotometer; and (4) checking the content of the glucuronic acid in the sample tube according to the
absorbance of the sample tube from a
standard curve, and then calculating the content of the free
sodium hyaluronate according to a calculation formula. The method disclosed by the invention is used for detecting the content of the free
sodium hyaluronate, the experimental results are stable and reliable, and the
repeatability is good.
