The palladium-based dual functional film contains a palladium film or palladium alloy film deposited on the surface of porous ceramics and is characterized in that pore channels of the porous ceramics are loaded with a methanation catalyst. The preparation method comprises steps of: loading the methanation catalyst into the pore channels of the porous ceramics by an immersion method and preparing the palladium or palladium alloy film on the surface of the porous ceramics by a chemical plating or chemical plating and electroplating coupling method. According to the palladium-based dual functional film, the methanation catalyst is loaded in the pore channels of the palladium film or palladium alloy film porous carrier so as to realize the coupling of film separation and carbon monoxide catalytic methanation. When the film is used for hydrogen separation and purification, trace carbon monoxide and carbon dioxide leaked from defects of the film can undergo catalytic methanation, thus satisfying special hydrogen requirements. The method comprises the following steps: (1) strain activation: slant culture medium is inoculated with Enterobacter mori VL4-3 to obtain a slant culture, then a seed culture medium is inoculated with the slant culture, and transformation is conducted to obtain a seed culture solution; (2) complex biotransformation pretreatment: in the reconstituted tobacco extraction process, a reconstituted tobacco aqueous solution is inoculated with the seed culture solution, a compound enzyme of clostridium butylicum feruloyl esterase, lignin peroxidase (Lip) and manganese peroxidase (MnP) is added, uniform stirring and mixing are performed, and fermentation is conducted to obtain a reconstituted tobacco biotransformation solution; (3) reconstituted tobacco is produced through a conventional process of reconstituted tobacco, wherein in the process, the temperature exceeds 75 DEG C, enzymes and the bacteria are inactivated, and rolling is conducted to obtain the reconstituted tobacco. According to the reconstituted tobacco, of which the fragrance quality is improved by the biotechnology, produced by the method, the irritation is reduced, the green miscellaneous gas is reduced, the fragrance is rich and elegant and is sweet as milk, and the fragrance quality of the reconstituted tobacco is improved.

The comprehensive method comprises the following steps: (1) compound fermentation: in a reconstituted tobacco extracting process, fermenting a reconstituted tobacco water solution with a compound of aspergillus niger, feruloyl esterase, soil lignin peroxidase, bacillus licheniformis powder and freeze-dried lactobacillus powder; (2) adsorbing nicotine in an extracting solution by virtue of strong acid in-exchange macroporous resin for 30-60min, and conducting filtering to remove the nicotine by virtue of a sieve so as to obtain an extracting solution; (3) concentrating the reconstituted tobacco in a vacuum environment at 75-85 DEG C until a solid content is 40%, so as to obtain a concentrated solution; (4) drying residues, which are obtained by extracting the reconstituted tobacco, by virtue of a dryer at 100-120 DEG C, and deactivating enzyme and bacteria so as to obtain tobacco substrates; and (5) rolling the tobacco substrates. With the application of the comprehensive method provided by the invention, the produced reconstituted tobacco is low in irritation, mellow in taste and smell, elegant in aroma, light in green miscellaneous gas and smooth in smoke, and the all-around improvement of the quality of the reconstituted tobacco is achieved.
