The invention provides a microRNA quantitative detection analytic method by utilizing isothermal amplification to synthesize a fluorescent nano silver cluster probe. The method is as below: a DNA amplification template containing three kinds of functional sequences is designed: a sequence binding with a target microRNA, a nicking endonuclease enzyme sequence and a DNA complementary sequence for synthesis of fluorescent nano cluster; when the target microRNA and the DNA amplification template are combined, isothermal amplification reaction and specific enzyme reaction of nicking endonuclease are induced to obtain a large number of single-stranded DNA products; the DNA sequence for synthesis of fluorescent nano silver cluster and a silver nitrate solution are employed to prepare the fluorescent nano silver cluster probe under the reduction of sodium borohydride; fluorescence signal of the reaction system are determined, and a fluorescence change value is calculated; the value is compared with a standard working curve to calculate the concentration of the target microRNA. The method has the advantages of high sensitivity, strong specificity, wide linear detection range, low background signal and simple operation, and can be widely applied to microRNA detection of biological samples such as tissue, blood or cells.

The invention discloses a rapid synthesis method of an indium phosphide polycrystalline material, which comprises the following steps of: (I) performing surface cleaning treatment on indium; (II) placing a multi-tubular quartz phosphorus bubble with phosphorus into a phosphorus source furnace; (III) placing the pre-designed thermal insulation system, heater, crucible with indium, phosphorus source furnace and seed crystal and B2O3 into a high-pressure single-crystal furnace hearth; (IV) vacuumizing the inside of the furnace and filling high-purity argon; (V) heating to vaporize phosphorus in the phosphorus bubble and injecting into indium melt, and reacting to generate indium phosphide; and (VI) growing crystal. The multi-tubular quartz phosphorus bubble comprises a quartz phosphorus container, at least two quartz phosphorus bubble tubes and a quartz cover. By adopting a multi-tubular quartz phosphorus bubble, the method disclosed by the invention realizes a rapid multi-tubular phosphorus injection synthesis technology, solves the problems of long synthesis time, non-uniform melt proportioning, impurity pollution and the like in the original technology, realizes rapid, efficient and high-purity synthesis of the indium phosphide material, and performs InP single crystal growth more easily.

The invention discloses a cultivation method for inducing scutellaria baicalensis hairy root based on agrobacterium rhizogenes infection, which uses an agrobacterium rhizogenes bacteria liquid to infect root of a scutellaria baicalensis explant and induces the scutellaria baicalensis explant to form a hairy root, the scutellaria baicalensis hairy root presents yellow hairy on a MS solid medium, so that scutelloside can be stably synthesized, an ITS sequencing is performed and the hairy root can be identified as the scutellaria baicalensis hairy root of a scutellaria. Then seed selection is performed for further in a seed selection medium and a cultivation condition can be optimized to select lines which raise the content of active ingredient scutelloside used for hairy root traditional Chinese medicines. Hairy root can be obtained through scutellaria of a scutellaria introduced by agrobacterium rhizogenes, wherein the inductivity can reach 10 - 40%, the obtained scutellaria baicalensis hairy root presents yellow hairy which is capable of stably synthesizing scutelloside. According to the invention, the scutellaria baicalensis hairy root of a scutellaria is cultivated on the optimized medium, the growth is rapid and the biomass can be increased for 23 -35 times in 30 - 35 days, and the cultivation is performed under the dark conditions. The invention has the characteristics of low energy consumption and low cost, which is suitable for industrialization production.

Synthetic process for gem grade diamond

A synthetic process for gem grade diamond comprises the following steps: (1) placing an assembled synthetic block in a high pressure chamber of a cubic press; after pressurizing the synthetic block to a final pressure of 90-95 MPa at one time, maintaining the pressure; when the pressure rises to 40-50% of the final pressure, starting heating and enabling the heating power to rise to 4-5 kw within 10-15 s; after preserving the heat of the synthetic block for 3-5 min, increasing the heating power to 9-10 kw for heat preservation; (2) after preserving the heat of the synthetic block for 5-10 h, entering a continuous heat preserving and pressure maintaining stage after continuous temperature increasing, heat preserving, pressure increasing and pressure maintaining; (3) after continuous heat preservation, stopping heating and cooling the synthetic block to room temperature; releasing the pressure when the temperature is halved; finishing the process till the pressure is released to normal pressure. The synthetic process for the gem grade diamond is especially suitable for multi-crystal-seed synthetic processes, can reasonably supply enough carbon source according to the growing speeds of crystal seeds in the control process and can enable the crystal seeds to stably grow for a long time without being affected by spontaneous nucleation to synthesize large particle size and inclusion-free high-quality diamond single crystal; the crystal is colorless and transparent and the quality of the crystal is good; the synthetic process for the gem grade diamond can realize mass production and low production cost.
