The invention relates to the technical field of urea detection, and especially relates to a urea detection reagent. A reagent R1 contains an imidazole buffer solution, BSA, alpha-oxpentanedioic acid, sucrose, trehalose, GLDH, polyethylene glycol, dodecyltrimethylammonium chloride, lauryl imidazolinium betaine and an antiseptic; and a reagent R2 contains a Tris buffer solution, the BSA, urease, sucrose, trehalose, NADH and the antiseptic. The imidazole buffer solution is adopted, and the BSA, sucrose, trehalose, polyethylene glycol, xanthan gum and mannitol which are used as a stabilizer are added, so the stability of the reagent is greatly enhanced; and dodecyltrimethylammonium chloride and the lauryl imidazolinium betaine which are used as novel surfactants substantially improve the detection performance and enhances the stability of the reagent.
